dc.descriptionEl objetivo del presente trabajo de titulación fue evaluar mediante ensayos in vitro la actividad antioxidante y antiinflamatoria de los extractos hidroalcohólicos de hojas de Myrcianthes hallii (Arrayán). Una vez recolectado el material vegetal se secó y trituró. Para obtener los extractos, las hojas se maceraron con etanol al 70% v/v, y se concentró en evaporador rotatorio bajo condiciones controladas, para posteriormente liofilizar los residuos. Los flavonoides espectrofotométricamente a través del método del Tricloruro de Aluminio, obteniendo los siguientes resultados: 11646,095 ± 125,367 mg Eq Q/100 mg de extracto liofilizado de hojas. La determinación de fenoles se realizó por el método de Folin-Ciocalteu dando el resultado de 45633,33 ± 577,35 mg Eq GAE/100 mg de extracto liofilizado de hojas. El extracto presentó una capacidad captadora de radicales libres a un IC50 de 102,45 ug/ml con un rango de 87,67 a 119,72 ug/ml para las hojas, el cual fue evaluado por el método de DPPH˙. El ensayo de actividad antiinflamatoria se llevó a cabo por el método in vitro de neutrófilos aislados, por medio de la determinación de una sal de tetrazolio estable (WST-1). Las hojas de Myrcianthes hallii, presentaron actividad antiinflamatoria a una IC50 de 31,68 ug/ml con un rango que va de 21,23 – 46,19 ug/ml. La concentración de 200 ppm presentó el mejor porcentaje de inhibición inflamatoria para hojas con 79,67 ± 3,02 %. Estos porcentajes son mayores en comparación con el ácido acetilsalicílico usado como referencia que a 200 ppm presentó 68,88 ± 3,90 %. Los ensayos se realizaron por triplicado. Se concluyó en base al control de calidad del extracto hidroalcohólico de las hojas de Myrcianthes hallii se determinaron factores como: requisitos organolépticos, densidad relativa, índice de refracción, sólidos totales y pH, dando resultados característicos de la especie y del solvente usado, por lo que se recomienda realizar ensayos sobre la actividad cicatrizante, antioxidante y antiinflamatoria haciendo uso de aceites esenciales y de la porción alcaloidal de las hojas de Myrcianthes hallii.es_ES
dc.description.abstractThe objective of the fallowing investigation was to evaluate the antioxidant and anti-inflammatory effects of the water-alcohol extracts of the Myrcianthes hallii (Arrayán) leaves using in vitro tests. Once the vegetable matter was collected, it was dried and ground. In order to obtain the extracts, the leaves were ground with 70% ethanol (v/v). Then, it was made more concentrated using a rotary evaporator under a controlled environment. After that, the refuse was freeze- dried. The flavonoids were processed using aluminum trichloride and a spectrophotometer, through which the fallowing results were obtained: 11646, 095 ± 125, 367 mg (equiv) Q/100 mg of freeze- dried extract of leaves. The phenols were determined using the Folin-Ciocalteu method, through which, 45633, 33 ± 577, 35 mg equiv GAE/ 100 mg of freeze- dried extract of leaves were obtained. The extract’s capacity of attracting free radicals at a half maximal inhibitory concentration (IC50 ) of 102, 45 mcg/ml with a range of 87, 67 to 119, 72 mcg /ml for the leaves was evaluated using the DPPH method. The anti-inflammatory trial was carried out using the in vitro of isolated neutrophils method. A stable tetrazolium chloride (cell proliferation reagent, wst-1) was used during this process. The leaves of Myrcianthes hallii had an anti-inflammatory effect at a half maximal inhibitory concentration (IC50 ) of 31,68 mcg /ml with a range from 21,23 to 46,19 mcg /ml. The 200-ppm concentration showed the best percentage of inflammatory inhibition for the leaves with 79, 67 ± 3,02 %. These percentages are greater in comparison with those of the acetylsalicylic acid (aspirin), using as a refence, that at 200-ppm showed 68,88 ± 3,90 %. The trials were carried out three times. The following factors were determined, based on the quality control of the water-alcohol extracts of the Myrcianthes hallii: organoleptic requirements, relative density, refraction index, total solids and ph. The results obtained were characteristic of this specie and the solvent used. For these reasons the authors recommend to do clinical trials in order to evaluate the healing, antioxidant and antiinflammatory effects using the essential oils and the alkaloidal portion of the Myrcianthes hallii leaves.es_ES
